We've had people take one-way rentals to and from Portland, ME when we're in Mount Desert Island.

Check that out before you you tie yourself to some other place in Maine.
We've found (in the past going West), that Southwest Harbor is a good location. You could spend a month or more just exploring west of there in Penobscot Bay.
Beyond that, you can make an workable jumps down the coast -- or take an overnight hop down the Marblehead, Provincetown, etc.
